    WARP: A ICN architecture for social data

    Social network companies maintain complete visibility and ownership of the data they store. However users should be able to maintain full control over their content. For this purpose, we propose WARP, an architecture based upon Information-Centric Networking (ICN) designs, which expands the scope of the ICN architecture beyond media distribution, to provide data control in social networks. The benefit of our solution lies in the lightweight nature of the protocol and in its layered design. With WARP, data distribution and access policies are enforced on the user side. Data can still be replicated in an ICN fashion but we introduce control channels, named \textit{thread updates}, which ensures that the access to the data is always updated to the latest control policy. WARP decentralizes the social network but still offers APIs so that social network providers can build products and business models on top of WARP. Social applications run directly on the user's device and store their data on the user's \textit{butler} that takes care of encryption and distribution. Moreover, users can still rely on third parties to have high-availability without renouncing their privacy

    The genetic study of three population microisolates in South Tyrol (MICROS): study design and epidemiological perspectives

    <p>Abstract</p> <p>Background</p> <p>There is increasing evidence of the important role that small, isolated populations could play in finding genes involved in the etiology of diseases. For historical and political reasons, South Tyrol, the northern most Italian region, includes several villages of small dimensions which remained isolated over the centuries.</p> <p>Methods</p> <p>The MICROS study is a population-based survey on three small, isolated villages, characterized by: old settlement; small number of founders; high endogamy rates; slow/null population expansion. During the stage-1 (2002/03) genealogical data, screening questionnaires, clinical measurements, blood and urine samples, and DNA were collected for 1175 adult volunteers. Stage-2, concerning trait diagnoses, linkage analysis and association studies, is ongoing. The selection of the traits is being driven by expert clinicians. Preliminary, descriptive statistics were obtained. Power simulations for finding linkage on a quantitative trait locus (QTL) were undertaken.</p> <p>Results</p> <p>Starting from participants, genealogies were reconstructed for 50,037 subjects, going back to the early 1600s. Within the last five generations, subjects were clustered in one pedigree of 7049 subjects plus 178 smaller pedigrees (3 to 85 subjects each). A significant probability of familial clustering was assessed for many traits, especially among the cardiovascular, neurological and respiratory traits. Simulations showed that the MICROS pedigree has a substantial power to detect a LOD score ≄ 3 when the QTL specific heritability is ≄ 20%.</p> <p>Conclusion</p> <p>The MICROS study is an extensive, ongoing, two-stage survey aimed at characterizing the genetic epidemiology of Mendelian and complex diseases. Our approach, involving different scientific disciplines, is an advantageous strategy to define and to study population isolates. The isolation of the Alpine populations, together with the extensive data collected so far, make the MICROS study a powerful resource for the study of diseases in many fields of medicine. Recent successes and simulation studies give us confidence that our pedigrees can be valuable both in finding new candidates loci and to confirm existing candidate genes.</p

    Association of kidney disease measures with risk of renal function worsening in patients with type 1 diabetes

    Background: Albuminuria has been classically considered a marker of kidney damage progression in diabetic patients and it is routinely assessed to monitor kidney function. However, the role of a mild GFR reduction on the development of stage 653 CKD has been less explored in type 1 diabetes mellitus (T1DM) patients. Aim of the present study was to evaluate the prognostic role of kidney disease measures, namely albuminuria and reduced GFR, on the development of stage 653 CKD in a large cohort of patients affected by T1DM. Methods: A total of 4284 patients affected by T1DM followed-up at 76 diabetes centers participating to the Italian Association of Clinical Diabetologists (Associazione Medici Diabetologi, AMD) initiative constitutes the study population. Urinary albumin excretion (ACR) and estimated GFR (eGFR) were retrieved and analyzed. The incidence of stage 653 CKD (eGFR &lt; 60 mL/min/1.73 m2) or eGFR reduction &gt; 30% from baseline was evaluated. Results: The mean estimated GFR was 98 \ub1 17 mL/min/1.73m2 and the proportion of patients with albuminuria was 15.3% (n = 654) at baseline. About 8% (n = 337) of patients developed one of the two renal endpoints during the 4-year follow-up period. Age, albuminuria (micro or macro) and baseline eGFR &lt; 90 ml/min/m2 were independent risk factors for stage 653 CKD and renal function worsening. When compared to patients with eGFR &gt; 90 ml/min/1.73m2 and normoalbuminuria, those with albuminuria at baseline had a 1.69 greater risk of reaching stage 3 CKD, while patients with mild eGFR reduction (i.e. eGFR between 90 and 60 mL/min/1.73 m2) show a 3.81 greater risk that rose to 8.24 for those patients with albuminuria and mild eGFR reduction at baseline. Conclusions: Albuminuria and eGFR reduction represent independent risk factors for incident stage 653 CKD in T1DM patients.     Overexpression of the Cytokine BAFF and Autoimmunity Risk

    BACKGROUND\textbf{BACKGROUND}: Genomewide association studies of autoimmune diseases have mapped hundreds of susceptibility regions in the genome. However, only for a few association signals has the causal gene been identified, and for even fewer have the causal variant and underlying mechanism been defined. Coincident associations of DNA variants affecting both the risk of autoimmune disease and quantitative immune variables provide an informative route to explore disease mechanisms and drug-targetable pathways. METHODS\textbf{METHODS}: Using case-control samples from Sardinia, Italy, we performed a genomewide association study in multiple sclerosis followed by TNFSF13B locus-specific association testing in systemic lupus erythematosus (SLE). Extensive phenotyping of quantitative immune variables, sequence-based fine mapping, cross-population and cross-phenotype analyses, and gene-expression studies were used to identify the causal variant and elucidate its mechanism of action. Signatures of positive selection were also investigated. RESULTS\textbf{RESULTS}: A variant in TNFSF13B, encoding the cytokine and drug target B-cell activating factor (BAFF), was associated with multiple sclerosis as well as SLE. The disease-risk allele was also associated with up-regulated humoral immunity through increased levels of soluble BAFF, B lymphocytes, and immunoglobulins. The causal variant was identified: an insertion-deletion variant, GCTGT→A (in which A is the risk allele), yielded a shorter transcript that escaped microRNA inhibition and increased production of soluble BAFF, which in turn up-regulated humoral immunity. Population genetic signatures indicated that this autoimmunity variant has been evolutionarily advantageous, most likely by augmenting resistance to malaria. CONCLUSIONS\textbf{CONCLUSIONS}: A TNFSF13B variant was associated with multiple sclerosis and SLE, and its effects were clarified at the population, cellular, and molecular levels.     Population- and individual-specific regulatory variation in Sardinia

    Genetic studies of complex traits have mainly identified associations with noncoding variants. To further determine the contribution of regulatory variation, we combined whole-genome and transcriptome data for 624 individuals from Sardinia to identify common and rare variants that influence gene expression and splicing. We identified 21,183 expression quantitative trait loci (eQTLs) and 6,768 splicing quantitative trait loci (sQTLs), including 619 new QTLs. We identified high-frequency QTLs and found evidence of selection near genes involved in malarial resistance and increased multiple sclerosis risk, reflecting the epidemiological history of Sardinia. Using family relationships, we identified 809 segregating expression outliers (median z score of 2.97), averaging 13.3 genes per individual. Outlier genes were enriched for proximal rare variants, providing a new approach to study large-effect regulatory variants and their relevance to traits.     Reliable Content Distribution using ICN

    Information Centric Networking [ADI12] has an intrinsic distributed nature, which eases decentralizing formerly centralized protocols. Nevertheless, implementing fully distributed protocols on a large scale remains challenging due to the concurrent nature of network elements.Cache inconsistency becomes a problem whenever different content, or multiple versions of the same content, can answer the same query, in the first part of this book we focus on the detrimental impact that this has on Privacy and Communication Many-To-Many.With regard to privacy we show that hierarchical caching introduces a considerable la- tency in updating Access Control Lists. We study the problem analytically and via simula- tion and propose two possible solutions: one for the generic problem of privacy in ICN and one for the specific case of Social Networking.In the latter case of many-to-many communication, we prove that the current state of the art limits the throughput and frequently suffers data loss.The second and last part of the book switches topic to the impact of ICN on wireless ad-hoc networks.     BLOOGO: BLOOm filter based GOssip algorithm for wireless NDN

    Modern ad-hoc networks focus on data rather than ob- solete end-to-end communications. This paper describes BlooGo, a gossip algorithm that disseminates messages throughout the network with a minimum number of trans- missions. Its peculiarity lies on the fact that without any knowledge of the network the receiver of a packet can de- cides autonomously whether to forward it or not. All the information that BlooGo uses to decide the utility of a transmission is enclosed in the packet as a bloom filter; this way the parties, i.e. sender and receiver, never have to communicate directly. This approach makes BlooGo state-less; lightweight; power-efficient; ideal for sensors or embedded devices that use the NDN philosophy. \uc2\ua9 2012 ACM

    MADN - Multipath Ad-hoc data network prototype and experiments

    This paper presents the first prototype of Multipath Ad-hoc Data Network (MADN) a clean slate protocol for ad-hoc wireless content distribution. Inspired by the recent advances on Information Centric Networking (ICN), MADN is namely a pull-based protocol that emphasizes on seamless route redundancy and multi-path data delivery. It is complementary to the former IP protocol and therefore does not implement primitives for machine-to-machine communication. In consideration of targeting specifically the wireless medium, it uses rateless encoded data packets to counterbalance packet loss and to benefit from multi-source data distribution. A major difference between MADN and previous works is the use of the BlooGo algorithm in place of the conventional routing protocols - e.g. random walks, landmarks or coordinate spaces. The main advantages of using BlooGo is that it delivers the packets in considerably less hops while it maintains alternative routes open as backup of the shortest ones. This paper serves two aims, firstly it proposes a new protocol for multipath content distribution in ad-hoc environments, secondly it presents its architecture and how this interfaces with the final applications. Ultimately, the implementation is intentionally kept modular in order to facilitate deploying and evaluation of new solutions and new configurations - e.g. different caching algorithms or different types of Erasure Codes - without major refactoring of the codebase. \uc2\ua9 2013 IEEE

    In this paper we conduct a performance evaluation of privacy protocols for Information Centric Networking (ICN). Our contribution is three-fold: Firstly, we define a simple but complete performance framework for comparing current and future solutions. Secondly, we conjecture and prove the existence of unsafe replicas, namely cached content that remains available to users whose access has been revoked. Thirdly, we propose a performant protocol that solves the problem of unsafe replicas without tampering with the caching functionality of ICN
